In order to solve the startup display problem of Vitrobot 3 (firstly found on December 06, 2024), an upgrade kit was installed by the engineer on February 20, and the machine resumed service after actual tests. Attached within please find the reminders and comparisons between the previous and the current versions of the operating interface. PDF
In addition, the CEM3 sample cassette gripper malfunctioned again on the afternoon of February 14, but the condition was determined as good after the inspection and the multiple tests conducted by the engineer on February 17, so the cause remains to be confirmed; meanwhile, the Y axis measurement module was replaced during February 17-19 to solve the instability when running CEM3 Altas function. CEM3 is currently providing service as usual. Thank you!

The sample cassette gripper of CEM3 malfunctioned again in the afternoon on February 14, but was determined in good condition when the engineer checked it on February 17, so the cause remains to be determined. Afterwards, the Atlas Y axis measurement module and the Select Area (SA) AAM were replaced during February 17-19. CEM3 resumed service on February 20.

1. ASCEM has new web address https://cryoem.project.sinica.edu.tw
2. The calculation basis for the staff-operation charge will be raised to NT$600 per hour. The Vitrobot regulation is revised as below:
(1) Vitrobot (including training and practicing) charge rate is raised to NT$600 per unit (2 hours) for academic users and NT$2,400 for industry users ; if staff-operation is needed, the new charge rate is NT$1,200 per time.
(2) Non-AS user may operate Vitrobot after being trained and certified. The open hours are 09:00-16:00, Monday through Friday, and 2 hours per unit. User may borrow a temporary access card from ASCEM by a personal ID card to enter and exit the lab during the machine day.
(3) [3-Month Trial] Vitrobot user who book the session of 18:00-20:00 may borrow a temporary access card from ASCEM during office hours by a personal ID card to enter and exit the lab after 18:00 if necessary. User who operates instruments during off-hours is responsible for the maintenance of the entire facility and environment, his or her lab will be held for reparation responsibility for facility damages due to negligence of instructions.

CEM3 stopped functioning at 7pm on November 8 due to the unstable vacuum state caused by the failure of its air compressor. On November 11, the air compressor was restored after replacing the aged and damaged connecting pipe by the engineer, and so did the CEM3 vacuum state return to normal. On November 12, CEM3 is back in service. |

The CEM3 automatic liquid nitrogen filling system broken down at night, causing the machine to automatically return to temperature. After the inspection and logbook analysis by the ThermoFisher engineer, it was considered that the cause was non-human, and the machine operation has been being stable after manually filling liquid nitrogen on 11/7. |

Because we have received several inquiries to confirm the ASCEM service charge rates, to clarify the recent adjustments, this is to notify you that all academia users, no matter it is AS or non-AS budgets being used to pay, share the same charge scheme and discount from August 1, 2023. The charge rate for industry users currently remains the same as before. In addition, ASCEM provides three modes of technical support: basic, advanced, and collaboration (technical R&D). Training courses for machine operation and data computing analysis are also feasible after discussion. We would like to have your understanding for the two things below,
1. Recently, the facility water cooling system was found errors few times, so were the chillers of CEM3 and
CEM4 shortly experiencing failures one after another, and the overheating of the weather was reasonably suspected as the main reason. Although the whole cooling system was repaired soon, in response to the breakdown, we still had to urgently re-schedule some users’ machine time then, and thank you very much for your cooperation and flexibility about the unexpected events. To try to decrease the errors happening again in the future, the environment improvement for the outer water cooling system will also be carried out by the manufacturer in the near future.
2. Please be reminded that all users have to follow the facility service regulations, especially the new
members from labs are highly recommended to directly contact facility staff for any big or tiny concerns because improper operation of machines could cause a forced interruption of service and further affect other users’ right to use.

This is to announce that according to the conclusion from the ASCEM User Committee Meeting held on March 15, 2021, ASCEM now provides staff-operation for cryo-sample preparing by Vitrobot in order to fulfill the needs from AS and non-AS users. Please see “CryoSample/Data” webpage for the new regulations. However, considering the efficiency for the experiment going and the limited manpower of ASCEM, Vitrobot certification is still strongly encouraged for long-term users within AS. In addition, please do follow the reservation regulations for the rights of usage. Every case application needs to be approved by the Lab Supervisor/PI, or the case will be considered as “cancelled” if the Lab Supervisor/PI doesn’t complete the online approval procedures before the deadline. |

According to the facility cost analysis formula of Division of Instrument Service of Academia Sinica (AS), the cost per day is respectively NTD100,000 and NTD140,000 for CEM3 and CEM4. To promote the research and the application of cryoEM, ASCEM has been giving CEM3/4 academia users 90% discount of service charges since operation started under the significant financial subsidization from AS and Taiwan Protein Project (TPP). However, in response to the new policy of AS and the budget reduction of TPP since 2020, ASCEM has to adjust service rate of CEM3/4. While the payment using AS budgets remains the 90% discount of service charges as before, using non-AS budgets is adjusted to 65% off. Meanwhile, considering that the off-AS researchers usually are funded by non-AS budgets only and may lack of cryoEM professional manpower in time or have limited budgets and so on, TPP has developed a cryoEM platform with a professional research team for supporting those in need to apply and take advantage of the resource. |
