 *The instruments of CLEM for cryo-fluorescence and of FIB for milling are external equipment that are respectively managed
   by the Institute of Plant and Microbial Biology (IPMB) and the Research Center for Applied Sciences (RCAS), Academia
   Sinica. Because they are not open to users outside the institutes, and must be equipped with special accessories for
   biological experiments, please contact ASCEM first if necessary.
Transmission Electron Microscopes (TEM)
 
CEM1 - JEOL JEM-1400

 
Room temperature TEM. Max. 120kV; Gatan CCD camera. Easy for operation, high quality images, can work as a cryo-EM as well.

 
Mainly for negative staining, the first step of checking the quality of protein or particle samples in the total cryo-EM workflow. Applicable to low-contrast sample observation in medicine, biology, clinics, immunology, pharmaceuticals, food, polymers etc.

CEM2 - FEI Tecnai G2 F20 TWIN
Max. 200k; Gatan 4K X 4K CCD and DE20 high-sensitivity camera.

 
Mainly for biological microstructure analysis, preliminary study of cryo-EM or cryo-tomography, sample screening and a low-resolution model building.
Also supports other studies, such as liposome and polymer etc.

CEM3 - FEI Talos Arctica 200 keV
Falcon 3EC detector; Volta phase plates; multi-purpose.

 
 
Mainly for automated condition screening and publication quality studies of both cryo-EM and cryo-TM, including overflow of the preliminary study samples from CEM2, screening of grids for CEM4, data collection of high quality data for structural determination (up to 4 angstrom or a little better) and publication.

Mainly for data collection for high-resolution structures. Preliminary sample, with biological significance, checking by CEM1/2/3 is recommended.

Model 950. Superior cleaning recipes and ratio of H2/O2/Ar.

 
Mainly for TEM samples, e.g. holey carbon grids, cleaning, remove hydrocarbon contamination. Supports grids preparation for SPA and cryo-tomography experiments.
 
PELCO easiGlow 91000 Glow Discharge System
Model 91000. Dual chamber to support hydrophilic and hydrophobic surface treatments separately to avoid cross contamination of chambers.

 
Mainly for TEM grids cleaning and carbon support films hydrophilic surface treatments. Supports grids preparation for SPA and cryo-tomography experiments.
 
Jelight UVO-cleaner
MODEL 18.
Mainly for cryo-grids cleaning prior to CEM3/4 data collection, removing organic contaminants.
 
FEI Vitrobot

 
Type IV。Instant cool bio-samples to the temperature of liquid nitrogen in less than microseconds, and freeze bio-molecules in very thin amorphous ice.

 
Mainly for SPA and cryo-Tomography experiments. Also supports other studies such as liposome and polymer, etc.
Computing System
 
ASCEM cooperates with the Core Facility of Big Data Analysis & Scientific Computing Academia Sinica (formerly ASGC) to construct a user-based integrated system of data transfer and process to cope with the huge amount of data of cryo-EM experiments. Services provided by ASGC will follow ASGC regulations, including charging schemes.
